Key Factors Driving the Growth of the Prader-Willi Syndrome Market

  • Rising Prader-Willi Syndrome Prevalence: As per DelveInsight’s analysis, in 2025, the US accounted for approximately 12,200 diagnosed prevalent cases of PWS. These are expected to increase during the forecast period (2026−2036) owing to improved screening methods leading to earlier detection, changes in lifestyle and environmental factors, and advancements in medical technology allowing for better diagnosis and treatment.
  • Growing Demand for Effective Treatments: PWS is associated with hyperphagia, obesity, growth abnormalities, and endocrine complications, creating a significant need for therapies that address multiple disease manifestations. The limited availability of disease-modifying treatment options is encouraging continued therapeutic development.
  • Increasing Use of Growth Hormone Therapy: Growth hormone treatment plays an important role in managing growth failure and body-composition abnormalities associated with PWS. Continued clinical experience and wider recognition of its benefits are supporting demand for growth hormone-based management.
  • Advances in Genetic and Molecular Diagnostics: Improvements in molecular and genetic testing are enabling more accurate confirmation of PWS and differentiation from other causes of developmental and metabolic abnormalities. Earlier diagnosis can facilitate timely multidisciplinary management and long-term monitoring.
  • Launch of Emerging Prader-Willi Syndrome Medications: The dynamics of the Prader-Willi syndrome market are expected to change in the coming years due to the launch of emerging therapies such as WAKIX (Harmony Biosciences), ARD-101 (Aardvark Therapeutics), PBF-999 (Palobiofarma), CSTI-500 (ConSynance Therapeutics), HBS-102 (Harmony Biosciences), Setmelanotide (Rhythm Pharmaceuticals), BMB-101 and BMB-105 (Bright Minds Biosciences), TNX-2900 (Tonix Pharmaceuticals), EPM301 (epm Therapeutics), and others.

Aparna Thakur, Project Manager of Forecasting & Analytics at DelveInsight, said that the introduction of new therapies, particularly those with novel mechanisms of action or broader symptom coverage, may fragment prescriber preferences and intensify competition for patient share. This could pressure VYKAT XR’s pricing flexibility, market uptake, or label expansion strategies, particularly if newer entrants demonstrate superior efficacy, safety, or convenience.

Prader-Willi Syndrome Market Analysis

  • Treatment strategies for Prader-Willi syndrome are tailored to the specific symptoms experienced by each patient.
  • Growth hormone replacement therapy remains the only FDA-approved treatment for children with PWS; however, its indication is limited to addressing growth failure and does not target key manifestations such as hyperphagia or behavioral abnormalities.
  • The FDA has approved three growth hormone products for use in PWS: GENOTROPIN, NORDITROPIN, and OMNITROPE. By comparison, the EMA and MHLW have approved only GENOTROPIN for PWS. OMNITROPE is also authorized as a biosimilar for PWS in Europe.
  • In March 2025, the US Food and Drug Administration (FDA) approved VYKAT XR (diazoxide choline) extended-release tablets, formerly known as Diazoxide Choline Controlled Release (DCCR), for the treatment of hyperphagia in adults and children aged 4 years and older with PWS. Following regulatory approval, VYKAT XR was officially launched, with patient treatment initiated in April 2025.
  • The development of therapies targeting PWS-associated hyperphagia has faced several challenges. Alize Pharma’s AZP-531 demonstrated efficacy in a Phase II trial in 2016 but did not advance into later-stage development. In the same year, Zafgen’s beloranib achieved positive results in a Phase III trial but was subsequently discontinued because of safety concerns.
  • Several companies are currently developing potential therapies for PWS, including Harmony Biosciences, with WAKIX (pitolisant) and HBS-102; Aardvark Therapeutics, with ARD-101; Palobiofarma, with PBF-999; and ConSynance Therapeutics, with CSTI-500, among other industry participants.

Prader-Willi Syndrome Competitive Landscape

Some of the Prader-Willi syndrome medications under development include WAKIX (Harmony Biosciences), ARD-101 (Aardvark Therapeutics), PBF-999 (Palobiofarma), CSTI-500 (ConSynance Therapeutics), HBS-102 (Harmony Biosciences), Setmelanotide (Rhythm Pharmaceuticals), BMB-101 and BMB-105 (Bright Minds Biosciences), TNX-2900 (Tonix Pharmaceuticals), EPM301 (epm Therapeutics), and others.

Harmony Biosciences’ WAKIX (pitolisant) is a selective histamine H₃ receptor antagonist/inverse agonist. The FDA has approved WAKIX for the treatment of excessive daytime sleepiness (EDS) or cataplexy in adults with narcolepsy. Although pitolisant is not currently approved for patients with Prader-Willi syndrome (PWS), it is being investigated as a potential treatment for the condition. The U.S. FDA has granted pitolisant Orphan Drug designation for the treatment of PWS.

Aardvark Therapeutics’ ARD-101 is an investigational, orally administered small-molecule therapy being developed to address hyperphagia-related behaviors in patients with PWS. Its proposed mechanism involves stimulating gut-localized secretion of cholecystokinin (CCK) and glucagon-like peptide-1 (GLP-1), which may help reduce hyperphagia-related behaviors. ARD-101 is currently undergoing Phase III development in the HERO trial.

Palobiofarma’s PBF-999 is an investigational, innovative phosphodiesterase 10 (PDE10) inhibitor developed by Palobiofarma. Preclinical studies have demonstrated potent and selective inhibition of the PDE10 enzyme, which plays an important role in regulating cyclic nucleotide levels, key signaling molecules involved in brain function. A Phase II clinical trial is currently evaluating PBF-999 in patients with PWS. The compound has previously been assessed in three clinical studies, where an appetite-suppressing effect was observed. Based on these findings, Palobiofarma has initiated a new clinical study to evaluate the safety and efficacy of PBF-999 in reducing hyperphagia among adults with PWS.

ConSynance Therapeutics’ CSTI-500 is an investigational triple monoamine reuptake inhibitor (TRI) that targets serotonin, dopamine, and norepinephrine by inhibiting their respective transporters—SERT, DAT, and NET. These neurotransmitters play important roles in regulating behavior, appetite, feeding, and mood. CSTI-500 is an orally administered New Chemical Entity (NCE) designed to potentially restore key neurotransmitter balance in patients with PWS.

The anticipated launch of these emerging therapies are poised to transform the Prader-Willi syndrome market landscape in the coming years. As these cutting-edge therapies continue to mature and gain regulatory approval, they are expected to reshape the Prader-Willi syndrome market landscape, offering new standards of care and unlocking opportunities for medical innovation and economic growth.
